Dissecting the mechanism of RNA metabolism machineries using Cryo-EM

The metabolism of RNA in eukaryotic cells is highly regulated via multiple pathways such as RNA quality control and RNA interference. The machineries involved in these pathways play important roles in gene regulation and cellular responses to stimuli. We have been using cryo-electron microscopy (cryo-EM) as a major structural biology tool to elucidate the mechanism of the RNA exosome complex in RNA quality control and the Dicer protein in RNA interference pathways. I will describe our recent discoveries on the structure and mechanism of the two machineries in their transactions with RNA substrates. These results provide us new insights to the regulation of RNA metabolism.
